Prahlad was a great devotee of Lord Vishnu. But his father, King Hiranyakashipu, was a tyrant and he was trying to please Lord Brahma to gain immortality.
Hiranyakashipu was an enemy of Vishnu and was angry with Prahlad for being a devotee of Vishnu. And so, Hiranyakashipu asked his sister Holika to kill Prahlad. Holika had a boon to protect her. Lord Brahma gave her a divine shawl that protected her from fire, and so she sat on the burning pyre with Prahlad in her lap. But when Lord Vishnu came to know about the plan of the two, the divine shawl protected Prahlad instead of Holika.
Holi is also a celebration of the divine love between Lord Krishna and Radha. So, an elaborate Holi is played in Mathura and Vrindavan.
